The Blewbury trainer had saddled just two winners since taking over the licence from her father, Fulke.
But Beaver Patrol landed the valuable StanJamesUk. com Heritage Handicap at Newmarket under Mick Kinane.
And that came 45 minutes after Mujood won the Royal Sussex Regiment Stakes at Goodwood.
"I can't believe it," she said.
"We had a winner at Goodwood and I thought that was exciting.
"I've been struggling to hold on to Beaver Patrol at home, but I am absolutely stunned by that performance - it's wildly exciting."
Johnson-Houghton's instructions to Kinane meant that the jockey can take many of the plaudits for the success.
"I said to Mick - 'Make sure he's in a race and then the rest is up to you as you are a top jockey.'"
East Ilsley handler Hughie Morrison was another to enjoy Newmarket success when his four-year-old Supaseus landed the StanJamesUk. com Suffolk Heritage Handicap.
The 10-1 shot held on by a short head to beat the fast-finishing Pinpoint.
The Royal Hunt Cup back over a mile could now be his aim.
